Used to determine the hemolytic effect of a test compound. Rabbit reticulocytes are exposed to a ten (10) point, two-fold dilution series of test article to assess its hemolytic potential after incubation at 37˚C for a set period of time (single time point between 15 and 180 minutes selected by client). The assay detects hemoglobin release as an indicator of red blood cell lysis and can be used to prioritize compounds and/or i.v. dosing regimens for subsequent in vivo studies. The percent hemolysis at each test concentration is calculated using the following formula:
Percent hemolysis = 100 x [(absorbance of sample – absorbance of negative control) / (absorbance of positive control – absorbance of negative control)].
